Swapping out the DVD-RW drive in my SAM for a plain ordinary DVD drive resolved the problem. I installed U-Boot 1.3.1b and PCI update without a hitch after this.
I cannot begin to guess what caused the old drive to stop working in this way. I guess it will remain a mystery!

@Mrodfr
I've confirmed that my Optical drive is connected to the J23 connector and also that my hard disk is connected to the J20 connector, both of which I understand to be the correct connectors.
I did a quick test and swapped over the two SATA ports, plugging the DVD ROM drive into the port the HD was in and vice versa. This made no difference other than the DVD drive was listed first on the BIOS screen and the HD second (and also OS4.1 failed to boot, reverting to the floppy disk screen immediately following the Amiga OS4.1 boot image). I think this though rules out a fault with J23 port??
Entering the NVgetvar command in shell got me a load of output, the key lines (I think) were;
sii3114ide_conf=1002 sii3114ide_xfer=G000 menuboot_cmd=boota boot1=s4iicdrom boot2=s4sii boot3=
Something that may be relevant is what the BIOS screen reports, but only if a CD is in the DVD drive. After detailing IDE Device 0: OK and detailing the hard disk type, support and capacity figures, it states "ATAPI_Issue : (no DRQ) after sending ccb (1b) status 0xd0" It then goes on to detail my DVD drive maker, model etc. The above may be a red herring, but thought it worth mentioning!
I'm now considering swapping out the DVD drive for another one, but I'm 100% certain this one used to boot CDROM's (such as the OS4.1 CDROM) as I built the system myself rather than get OS4.1 pre-installed!

@Phantom
I simply burned the pci-mini-update.iso to CD using Roxio 2010 on my PC. No option in the Disk Image Burn options to make it bootable.
However, I have exactly the same issue with the OS4.1 install CDROM too which is bootable.

[FIXED] Getting SAM (non-flex) to boot from DVD Drive

I've had this problem for a while and it's only become an issue since the new PCI Update for Sam440ep and Sam440ep-flex was release, which I downloaded and put onto a CD.
Whenever I try to boot from DVD I get the error "AOS4 FLB FLB: no SLB found in any of the designated boot sources; returning to u-boot."
This happens regardless of what option I select in the U-Boot Boot Select Boot Device menu, i.e. SII-4 SATA DVD, SII SATA DVD or SII PATA CDROM.
This happens regardless of the CDROM too, including the Amiga OS4.1 install CDROM.
In the recent past I did open up the SAM and check that the DVD drive was plugged into the correct SATA port on the motherboard, but I guess I need to check again. However, before I do this can someone advise on the correct options needed to boot from CDROM and/or suggest any other things I need to check/rule out.
The DVD drive works perfectly in Amiga OS4.1 and also I do recall booting successfully from CDROM at least once, cos I needed to reinstall the OS at one point about a year or more ago.

And as soon as I put up this post I discovered the solution - forgot to select "Bootable" in HDTools for the System partition! doh!

Os3.9 boot problems on A1200

I've put an 8GB CF card into my A1200 (with 3.1 ROMS), booted up via the Emergency disk, used HD Tools to create 4 x 1.8GB partitions and formatted the first 2 partitions using FFS as System and Work respectively (the final 2 remain unformatted).
OS3.9 installed perfectly OK, however upon rebooting the A1200, the HD drive light flashes briefly but the "install a floppy disk" animation shows.
Booting from the emergency disk again shows both System and Work as valid hard disk partitions, so what is stopping the A1200 booting from them?
p.s. I have used a 4GB CF card OK in this A1200, but that came pre-installed with WB3.0.

@kolla
USB 2 please and ASAP.
I see no reason for USB 3 development as there is no Amiga hardware with USB 3 ports and unless someone knows any different, I don't believe the X1000 or SAM460 are going to have them.
Let's get USB2 so that we can fully utilise the hardware in our current Amiga systems that is currently crippled by USB 1 only drivers.

@RIBDEVIL
When I try to use any of the themes I get the prompt "Please insert volume Datos: in any drive Process "<<IPrefs>>" followed by "Please insert volume Sistema: in any drive Process "<<IPrefs>>"
If I assign these to say my Data: and System: drives the screen refreshes as if the theme is applied, but doesn't look anything like the theme at all as the windows etc look very basic and I have to revert to the last saved theme to get the SilverGreen look back again.

A few questions about nice piece of kit which I've got on order.
I assume it'll work in the SAM (non flex) without issue and read and write to Amiga disks OK? However, does the Easy ADF software work ok on the SAM?
I got Easy ADF for use on my A1200, but wondered if it was SAM compatible. If Easy ADF does not work, is there another piece of software that works on SAM that'll enable me to create Amiga disks from ADF images?
My Windows PCs are the other option for use of the Catweasel, but alas the only two PC's I have are 64bit Windows 7, which I've seen threads discussing that the drivers don't work. Has anyone else got this to work on Windows 7 64bit?

@MickJT
Thanks for the tip MickJT. Alas that wasn't where I found the solution. In the end I spotted a directory called "Diskimagine" in Env-Archive and within that directory where two XML files that contained reference to SMBFS0:
With those two files removed the problems went away.

Startup error on my OS4.1 [SOLVED]

After playing around trying to install some classic apps software on my SAM from an old Amiga coverdisk, I've acquired a start up error message, which although I can bypass is a bit annoying;
"Amiga Dos Process: 78 Please insert volume SMBFS0: in any drive Process : "diskimagine.device.unit.0"
Any ideas how to stop this error? I presume the software put a command or something into my start up scripts, although I've checked Shell-Startup, User-Startup and Startup-sequence for anything new and cannot anything.
I can bypass the pop up by simply hitting Cancel, or assigning it to my Data: volume using the prompt's Assign button, but I'd rather it didn't appear at all.
Any tips on where to start looking would be appreciated!

Just posted this on AmigaWorld, but thought it best to post it here too Just back from the fair. To the others that attended - was it me or was the whole announcement thing badly organised? You could hardly hear the bloke from A-eon and couldn't see him either unless you were crammed at the front. At the very least he should have had a mic on, and/or had everyone stand in a horseshoe shape so more people could see him. In terms of info, the price will be "north of ?1,500" and they couldn't/wouldn't tell us the make of the CPU. They said it was likely the machine would be on sale before Xmas. It was cool to see the X1000 running OS4.1 though and the SAM 460 running. I'm very much inclined to go down the SAM 460 route now though, cos >?1,500 is a little steep.

Depending on the price difference between this and the X1000, this may be the system I buy if/when I decide to move on from my SAM, especially if there are graphics card drivers released for a good PCI-e card.

I got emails from AmigaKit at gone 11pm last night (they must work late!) saying they were enclosing my backplate free in with the order that I'd placed earlier yesterday (of BOH funny enough) and then another email canceling my backplate order and giving me a credit code for ?1.

Same has happened to me, a 0.48p charge. Trouble is my online banking has a minimum payment of ?1 (I normally pay via bank tranf) so I've overpaid them for the order.
What bugs me is less than an hour earlier I'd ordered something from them!!
